Neuroprotection of retinal ganglion cells with food materials -Supplement for prevention of glaucoma damage-

In Japan, glaucoma affects about 5% of people over 40 years old, and it is the most common eye disease causing blindness. In glaucoma, axonal lesions in the retinal ganglion cells cause visual field defects. Recent studies have implicated oxidative stress as a cause of retinal ganglion cell damage, and have reported that protecting the retinal ganglion cells with antioxidants is a useful treatment strategy. In this study, we focused on hesperidin, a food material with high antioxidant potential in primary cultures of mouse retinal cells. By administering hesperidin to the eyes of mice, we were able to reduce NMDA-induced retinal ganglion cell death and prevent the loss of vision. Furthermore, we determined that hesperidin has the antioxidant action, suppress calpain activation, an inducer of cell death, and that hesperidin acts as an anti-inflammatory. Hesperidin is a substance naturally found in large quantities in foods such as mandarin oranges, and supplements containing hesperidin promise to be useful for protecting against retinal ganglion cell damage in glaucoma. This research was conducted jointly by Professor Toru Nakazawa、Dr. Shigeto Maekawa and Dr. Kota Sato, of the Tohoku University Graduate School of Medicine, with the cooperation of Wakamoto Pharmaceutical Co., Ltd., and with the support of JSPS KAKENHI Grants-in-Aid for Scientific Research.
