OUTLINE

Most cancer cells have abnormal number of chromosomes (aneuploidy), which is caused by chromosomal instability, a condition in which chromosome missegregation occurs at a high rate. We are studying how chromosomal instability occurs in cancer cells, and found that alteration in chromosome dynamics during mitosis leads to chromosomal instability. Such alteration might be also related to aging. On the other hand, CHAMP1 (chromosome alignment-maintaining phosphoprotein), a molecule we discovered that is involved in chromosome segregation, was found to be also involved in DNA damage repair, and one of the responsible genes for intellectual disability. We are addressing these questions from the molecular to the organismal level using cultured cells and mice, and a variety of techniques including live-cell imaging, biochemical analysis, and genomic and epigenetic analyses. Through these approaches, we are aiming to develop a strategy to prevent disease by maintaining genome stability, and to develop cancer therapies by targeting chromosomal instability.

ほとんどのがん細胞では染色体数の異常(異数性)が認められ、その背景には染色体の不均等な分配が高頻度に起こる状態(染色体不安定性)が存在します。私たちはがん細胞における染色体不安定性の原因について研究を進めており、細胞分裂期の染色体の動態の違いが染色体不安定性につながることを見出しています。このような違いは、老化とも関係しているかもしれません。また私たちが染色体分配に関係する分子として発見したCHAMP1 (chromosome alignment-maintaining phosphoprotein)は、DNAの損傷修復にも関係しており、知的障害の原因遺伝子の一つであることが明らかになってきています。私たちはこれらのテーマについて、培養細胞やマウスを用いて、ライブセルイメージング、生化学的解析、ゲノム・エピゲノム解析などの手法を駆使することによって、分子レベルから個体レベルまでの一貫した理解を目指しています。そしてゲノムを安定に維持することによる疾患の予防や、染色体不安定性をターゲットとしたがん治療の開発につなげようとしています。
